Pancreatic cancer accounts for approximately 2.5% of all new tumors diagnosed worldwide, with more than 450,000 cases annually. Although its frequency is lower than that of other cancers such as breast or lung cancer, it is notable for its extreme aggressiveness and high mortality rate, making it one of the leading causes of cancer death globally. It has one of the lowest survival rates, as between 75% and 85% of patients are diagnosed at advanced stages due to the absence of initial symptoms.

To address this problem, a team of South Korean researchers has developed mussel-inspired nanoparticles that activate within cancerous tissue to destroy the tumor. This technology is inspired by mussels' ability to firmly adhere to rocks in the water. To achieve this, the research team created a protein similar to the one mussels use for adhesion using biotechnology. These proteins were then assembled into nanoparticles that carry gemcitabine, the standard chemotherapy drug for pancreatic cancer.

To allow them to travel safely through the bloodstream without being attacked by the immune system and without adhering to healthy tissue, the surface of the nanoparticles was coated with a layer of polyethylene glycol (PEG). The PEG shield was attached to the nanoparticle by a "hook" (a peptide) that is specifically cleaved by an enzyme called MMP2. This enzyme is produced in large quantities in pancreatic cancer tissue, but not in healthy tissue. The particle travels "camouflaged" by the PEG shield, without causing harm.

The PEG shield encounters the MMP2 enzyme, which cleaves and removes it. Without the shield, the nanoparticle regains its adhesive power and binds firmly to the tumor tissue, penetrating the cancer cells and delivering chemotherapy in a localized and continuous manner. Results in animal models of pancreatic cancer have been remarkable. Tumor volume and weight were reduced by half compared to the drug alone, and no systemic toxicity was observed, suggesting that the drug acts primarily on the tumor and does not harm healthy organs.
